(B) Myc-Hipk and HA-Arm plasmids were co-transfected into HEK293T cells. Lysates were incubated with anti-HA, anti-Myc or IgG (control) antibodies and immunoprecipitates were detected through WB with anti-HA or anti Myc, for Arm and Hipk, respectively. (C) Mammalian Hipk2 interacts with both β-catenin and Lef1. Flag-Hipk2 and T7-Lef1 or β-catenin were co-transfected into HeLa cells. Cell lysates were immunoprecipitated with indicated antibodies and protein complexes were visualized by immunoblotting with Flag, T7 and β-catenin. Hipk2 bound to T7-Lef1N and T7-Lef1C, deletion mutants that lack the β-catenin and HMG-binding domains, respectively.
