Y-specific PCR as described previously (B)

1996). (A) Multilineage reconstitution with 12 d.p.c. cells of
embryonic tissues was confirmed in some recipients by co-staining for donor
Ly5.1 and lineage-specific B220, Mac-1 and CD3
markers. In some
reconstituted mice the percentage of Ly5.1+, Mac- 1+ cells was very low (data
not shown). (B) Y-specific PCR analysis of blood samples. Each line on the gel
represents an individual mouse. The level of reconstitution was assessed with
reference to results of PCR using standard dilutions of male DNA in female
DNA. Arrowheads point to the mice reconstituted at the level of close to 10%
or more.
