(A) To determine whether HMG-box mutations altered the interaction between XSOX3 and b-catenin, HeLa cells were transfected with plasmids encoding XSOX3V5H6 alone or together with b-catenin and analyzed 24 hours later. Immunoblot analysis of cell lysates (?Lysate blots?) indicates that the exogenous polypeptides had accumulated to similar levels. Lysates were immunoprecipitated with anti-b-catenin and blotted with anti-V5; b-catenin co-immunoprecipitated both wild-type and mutant XSOX3 polypeptides. (B) Fertilized eggs were injected with RNAs encoding mutationally stabilized myc-tagged b-catenin either alone or together with RNAs encoding XSOX3. Lysates taken at stage 10 were analyzed by immunoblot (?Lysate blots?) to show that the exogenous polypeptides had accumulated to similar levels; b-catenin co-immunoprecipitated both wild-type and mutant XSOX3 polypeptides.
