Fig. S1. Effect of the raz mutation on 5¢ Hoxd expression. Whole-mount in situ hybridization of E10-12 forelimbs. Hoxd11 expression in E10.5 raz/raz embryos is expanded to include the anterior limb mesenchyme (compare C with A). Hoxd11 expression is severely downregulated at E11.5, but a light, thin rim of expression is noticeable in the distal mesenchyme (compare F with D). Hoxd12 expression is severely downregulated in raz/raz E12 forelimbs but, like Hoxd11, is expressed in a thin rim of distal mesenchyme (I). Hoxd13 is severely downregulated in raz/raz embryonic forelimb on E10.5 (L) and E12 (O), and at the latter stage is also expressed in a thin rim of distal mesenchyme.
Fig. S2. Expression of anterior restricted and posterior restricted genes in the raz mutant. (A-C) Pax9 expression on E11.5 in the left forelimb. Note the expression of Pax9 is restricted to the anterior mesenchyme of wild-type/wild-type and wild-type/raz embryos in A and B compared with the uniform A/P expression in the raz/raz embryo in C. (D-F) dHAND expression on E11.5 in the left forelimb. The expression of this gene is abundant in the posterior forelimb mesenchyme of wild-type/wild-type and wild-type/raz embryos (D,E), but is severely downregulated in the raz/raz forelimb (F).
