Fig. S1. The wgIL temperature sensitive allele cannot be used to determine the critical period of Wg action in promoting perimeter ommatidial PCD. (A) wgIL eye reared continuously at 16.5°C (the permissive temperature) for 112 hours APF (the equivalent of 42 hours APF at 25°C) showing a small amount of TUNEL stain (green). Many other wgIL eyes under these conditions showed less apoptosis (not shown). (B) wg eye reared at 16.5°C for 120 hours stained for Elav (red). Arrows indicate perimeter ommatidia that have not been eliminated. (C) Wild-type eye at 30 hours APF (at 25°C) showing many Wg-positive ommatidia. (D) wgIL eye at 16.5°C at an equivalent stage (80 hours APF) showing some ommatidia slightly positive for Wg (arrows). This is atypical: the vast majority of eye perimeters under these conditions have no Wg-positive ommatidia (not shown).
Fig. S2. The kinetics of apoptosis are slower in hidP mutants. (A-D) 42 hours APF hidP mutant eye stained for Wg (blue), TUNEL (green) and Elav (red). The onset of apoptosis in the ommatidia at the edge is delayed, as indicated by the presence of many cells positive for Elav, TUNEL and Wg, which is not seen in wild type (compare with Fig. 1E-H).
