Fig. S1. Unc-GFP does not exchange between centrioles and cytoplasm. Panels show centrioles in spermatocytes cultured from UNC-GFP-expressing adult testis, before and after photobleaching. GFP was bleached with a confocal laser, focused on one of a pair of centrioles (arrows); pairs were imaged before bleaching and at 1-2 minute intervals afterwards. Most unbleached centrioles retained a constant level of fluorescence over this time, but no fluorescence recovery was seen on the bleached centriole. Experiments were performed on cells 3-5 hours after cell dissection; cells retained normal morphology for 24-48 hours afterwards. Eight experiments were performed, all with similar results.
