Fig. S1. Validation of GFP;Hes1 and GFP;Notch1-IC lentiviral vectors. (A-F) Immunofluorescent staining demonstrating co-expression of GFP and Notch1-IC in COS7 cells infected with GFP;Notch1-IC, but not GFP alone. (G-L) Similar analysis demonstrating co-expression of GFP and Hes1 in COS-7 cells infected with GFP;Hes1, but not GFP alone. (M) Effective activation of a Notch/Su(H)-responsive luciferase reporter in COS-7 cells following either transfection with plasmid encoding myc-tagged Notch1-IC or infection with GFP;Notch1-IC lentivirus. Values indicate mean+s.e.m. for three independent conditions.
Fig. S2. Notch pathway activation in vimentin-positive mesenchymal elements in E15.5 mouse pancreas. Double immunostaining for Hes1 (green) and vimentin (red) confirms presence of nuclear Hes1 protein in vimentin-positive cells. At 15.5, widespread acinar cell differentiation has occurred, and little expression of Hes1 is observed in vimentin-negative epithelial cells.
