Fig. S2. Effect of TCE stem-loop III mutations on translational repression during oogenesis. Immunoblot analysis of HA-Nos protein in extracts of total ovary (T) and stage 14 egg chambers from nos-tub:TCEIIIGC/GC, nos-tub:TCEIIIA/U^C72 and nos-tub:TCE females using an anti-HA antibody. For each transgenic line, analysis of total ovarian extract confirms that the transgenic RNA is expressed and translated at earlier stages of oogenesis. Snf protein was monitored as a loading control. The TCEIIIGC/GC and TCEIIIA/U^C72 mutations partially compromise TCE function (Crucs et al., 2000), resulting in the appearance of HA-Nos in late oocytes.
