  Fig. S1. Sqt- and Cyc-expressing cells have the same properties as Tar*-expressing cells. Flat mounts of chimeric embryos at the shield stage showing the expression of tar, oep and ntl in blue (the probe is indicated in the bottom left of each panel). The progeny of transplanted cells expressing Sqt or Cyc is revealed by immunocytochemistry (red fluorescence staining the GFP). The genotype of the recipient embryo is labelled in the bottom right of each panel. +SU, embryos treated with 10 mM SU5402 after the transplantation. All views are of the animal pole.   Fig. S2. Endogenous oep expression is reduced but not abolished when fgf3 and fgf8 are inhibited. To investigate whether endogenous oep expression depends on the combined activity of Fgf3 and Fgf8, we assessed the expression of oep, tar and sprouty4 (sp4), which is a target of FGF signalling (Fürthauer et al., 2001), in wild-type embryos co-injected with fgf3 and fgf8 morpholinos. (A-R) Whole-mount embryos showing expression of tar, oep and sprouty4 in blue (bottom left) in uninjected wild-type embryos (uninj) or after injection at the one-cell stage of fgf3 and fgf8 morpholinos (MO3+8).
