Fig. S1. Ectopic expression of bcd at the posterior pole. (A) Whole-mount in situ hybridization to embryos from bcd-nos3'UTR females (referred to as bcd-nos3'UTR embryos) with a bcd-specific probe. Endogenous bcd mRNA is detected at the anterior and the hybrid bcd-nos3'UTR RNA in pole cells at the posterior. (B) Whole-mount antibody staining of bcd-nos3'UTR embryos with anti-Bcd monoclonal antibody detects the endogenous Bcd protein gradient in the anterior and ectopic Bcd in the pole cells. In some embryos, Bcd protein is also detected in somatic nuclei adjacent to the pole cells. Note that there is much less bcd-nos3'UTR RNA and Bcd protein at the posterior pole than endogenous bcd RNA and protein at the anterior. Embryos are oriented anterior towards the left and dorsal upwards.
