The wing pouch region of a hs-Delta, dpp-Gal4, UAS-D-mib animal was heat-shocked for 35 minutes, and then immediately dissected and stained for Myc-tagged D-mib (A, green) and Delta (B, red); merged image is shown in C. Fig. S2. Specificity of D-mib for internalization of DSL ligands. Experimental conditions are as described in Fig. 6M-O, except that discs were double-stained for Delta (green) and Notch (red). A lower field of magnification is shown in A-C than in D-I. The boxed region of B indicates the general focal field of D-I, which includes a characteristic central domain of elevated Notch expression that is marked with an asterisk. Approximate domains of ectopic D-mib expression, as marked by relocalized Delta, are indicated with white lines. Under conditions where large amounts of Delta are internalized by D-mib (D), Notch is not similarly internalized (E). In addition, degradation of Delta within the D-mib-expressing domain (G) is not accompanied by a comparable loss of Notch expression.
