Fig. S3. DNA content assay of cultured cells from the limb of
P1 mice. No significant difference was detected between day 1 (after
encapsulation) and day 21 (at harvest) in mutant and control cells. For DNA
assay, lyophilized hydrogel samples were homogenized in 1 ml papainase-PBE
solution [papain (125 g/ml, Worthington Biomedical, Lakewood, NJ), 100 mM
phosphate buffer, 10mM cystein, 10 mM EDTA, pH 6.3] and digested at 60°C for 16
hours. DNA content was determined by fluorophotometry with Hoechst 33258 dye
(Aldrich). Three samples were harvested for each assay.
