and ATH5 require E-boxes 2 and 4 to regulate the ATH5 promoter. Retinal cells
isolated at stage 29-30 (E6) were transfected with a wild type or a mutated
ATH5-promoter/CAT-reporter plasmid singly or in combinations with ATH5 or NGN2
expression vectors. Mutation of E-boxes 2 (mE2) and 4 (mE4) markedly reduced
promoter activity, whereas mutating the other E-boxes had little or no effect
(not shown). Overexpression of NGN2 or ATH5 did not activate the mutated
promoters.
