Movie 1. Cilia in Kupffer’s vesicle are motile. A dorsal view inside KV in a live embryo at the eight-somite stage8. Several cilia can be seen moving in this focal plane. Two of these cilia are circled. The elapsed time (seconds) is displayed in the lower right-hand corner.
Movie 2. Motile cilia in Kupffer’s vesicle generate a directional fluid flow. Fluorescent beads (red) were injected into KV at the six-somite stage and analyzed until the 12-somite stage. These beads showed a net counterclockwise flow inside KV. This directional flow is particularly apparent when the movie is sped up. This movie is an overlay of DIC and fluorescent images. Dorsal view with the notochord towards the right.
Movie 3. lrdr1 MO compromises fluid flow inside KV. Fluorescent beads (red) were injected into KV of lrdr1 morphant embryos (injected with lrdr1AUG MO at one- to four-cell stages). In this morphant, the counterclockwise flow inside KV observed in wild-type and control MO injected embryos was absent. Dorsal view with the notochord towards the right.
