1a and plexin 1 function in
guiding vulva cells within let-60(n1046gf) induced pseudovulvae. The smp-1p::gfp transcriptional reporter is expressed in dividing
P6.p cells in wild-type N2 animals (arrows in A). A similar smp-1p::gfp staining is observed in let-60(n1046gf) mutants, but in multiple vulva cell clusters (arrows
in B). In C (GFP fluorescence) and D (DIC), expression of the smp-1::gfp transcriptional reporter in let-60(n1046gf)
mutant pseudovulva cells (arrow) as they
form torroids. In a let-60(n1046gf) strain
carrying the ajm-1::GFP reporter,
we observe normal formation of the vulva torroids in the pseudo (E; white
arrow) and normal (right) vulvae, with the exception of the cell joining the
two vulvae. In plx-1(ev724);let-60(n1046gf) double mutants, we observe both normal (F; white
arrow) and abnormal (F-G; red arrows) vulva torroid development. The torroid
defects caused by the plx-1
mutation in pseudovulvae are similar to those we observe for the vulvae in a
non-Muv plx-1 background (see
Fig. 1). The perspective orientation for projections of 3D confocal images is
indicated with the triple arrows (D is for dorsal and P is for posterior; the
