Fig. S1. Lymphopoietic cells first emerge in the P-Sp/AGM area. A representative culture analysis of an E9.0-9.5 embryo is shown; this particular embryo had 19 somites. Tissues were dissected and placed as organ fragments on OP9 stromal cells as detailed in the Materials and methods. Cells were harvested after 2 weeks of culture and then subjected to flow cytometry analysis with the indicated markers.
Fig. S2. Lymphopoietic cells are present in the P-Sp/AGM area, but this activity is lacking in yolk sac hematopoietic cells. A representative experiment (the totals are summarized in Table 2) is shown where yolk sac and P-SP/AGM regions were dissected and dissociated before cell sorting. In this example, hematopoietic cell colonies were grown from E9.5 CD41+ Kit+ YS cells or Tie2+ P-Sp cells in OP9 stromal cell co-cultures. Upper panels illustrate clones visualized by phase contrast, while the lower ones show GFP fluorescence.
