Fig. S1. Analysis of CUC levels in wild-type, single and double mutants of cuc, syd and atbrm. Real-time RT-PCR was performed in duplicates using total RNA isolated from the above-ground tissues of 10-day-old seedlings. For double mutants, seedlings with fused cotyledons or petioles were analyzed. The ubiquitously expressed translation initiation factor EIF4A was amplified as an internal control. After normalizing with EIF4A, the resulting values for wild-type Ler were set to one to facilitate comparison between the lines. None of the single mutants shows a significant reduction in CUC expression, with the exception of cuc2-1. In this mutant, a transposon insertion in the CUC2 locus dramatically reduces expression of the gene. cuc1-1 syd-2 shows a reduction in CUC2 and CUC3 levels compared with the parental lines. cuc2-1 atbrm-1 shows a reduction of CUC1 and CUC3 expression levels compared with cuc2-1. cuc1-1 atbrm-1 has reduced CUC2 levels (not shown).
