Movie S1. Protrusive activity during neurulation in a wild-type embryo. Time-lapse movie (1 minute intervals, 2 hours and 51 minutes) of an mGFP-labeled embryo at the hindbrain level, imaged from a dorsal view (anterior towards the top), beginning approximately at 2-3 som and extending to 6-7 som. Superficial cells converge towards the midline by extending medio-lateral protrusive activity (arrow) and a deep cell extends protrusions towards the midline, while in contact with the lateral surface of the neural keel/rod (arrowhead).
Movie S2. Abnormal protrusive activity during neurulation in an Ncad-depleted embryo. Time-lapse movie (1 minute intervals, 2 hours and 24 minutes) of an mGFP-labeled, MO-injected embryo, at the hindbrain level, from a dorsal view (anterior towards the top), beginning approximately at 2-3 som and extending to 6-7 som. Cells appear to converge towards the midline only at the beginning of the movie. Later, cells remain in place sending out processes, however these extensions often retract, suggesting unstable protrusive activity.
Movie S3. β-cat-GFP distribution in wild-type cells during neurulation. Time-lapse movie (1.8 minute intervals on average, 2 hours and 34 minutes) of a β-cat-GFP-labeled embryo imaged from a dorsal view (anterior towards the bottom, midline towards the left) at the hindbrain level, beginning approximately at 2-3 som and extending to 6-7 som. The movie is a composite of a Z-stack (6 μm total). β-cat is not noticeably enhanced at the sites of cellular protrusions but rather appears to be distributed evenly throughout the plasma membrane.
