Fig. S1. Expression pattern of Ddok protein and mRNA in wild-type (wt) embryos of different
stages. (A-F) Ddok protein expression determined by immunofluorescence using
Ddok antiserum. Embryos were staged according to Campos-Ortega and Hartenstein
(Campos-Ortega and Hartenstein, 1997), and are oriented with anterior to left
and posterior to right and laterally viewed (except F, dorsolateral, and L,
dorsal). (A,B) Stage 2-3 (A) and stage 4-5 (B) are syncytial and cellular
blastoderm stages, respectively. Both of these stages show heavy maternal
contribution. (C,D,E) The stages of germband extension and retraction (stages
7-11), with ectodermal staining of Ddok. (F) Stages 12-14, when Ddok is
expressed in the hindgut (hg) and malphigian tubules (mpg). (G-L) mRNA
expression by whole mount in situ hybridization using a CO2H-terminal
Ddok antisense (digoxigenin-labeled)
riboprobe. The mRNA expression pattern is similar to the protein expression
pattern. Preimmune, preimmune antiserum staining of control stage 2-3 embryo.
Sense, Ddok sense riboprobe control,
stage 2-3 embryos. st, stomodeum; pr, proctodeum; cf, cephalic furrow; ep,
epidermis; pc, pole cells; am, anterior midgut; pm, posterior midgut; tr,
trachea.
