Fig. S1. Site 1 mediates
regulation of egl-1 in the P11
and P12 cell lineages. s.e.m. in all cases
was less than 10%. Transgenes included: wild type (a 7.6 kb genomic region
containing egl-1); 1,2 (mutation of
candidate sites 1 and 2 to NcoI sites);
3,4 (mutations of candidate sites 3 and 4 to NcoI sites); 2, 3 and 4 (individual mutations of candidate
sites 2, 3 and 4 to NcoI sites); and
1,2 F23D (a transgene containing NcoI mutations in sites 1 and 2 with deletion of a 370
nucleotide segment of the predicted phosphatase F23B12.1.) For the wild-type
data, 162 animals were scored from 11 independent lines. For the 1,2 transgene,
146 animals were scored from 10 independent lines. For the 3,4 transgene, 75
animals were scored from five independent lines. For the mutations affecting
Site 2 only, 75 animals from six independent lines were scored. For the
mutations affecting Site 3 and Site 4 only, 15 animals from one independent
line each were scored. For the Site 1,2 F23D 45, animals from three
independent lines were scored. The DXho transgene deletes the
leftmost 470 nucleotides of egl-1 from
the TRA-1 site (Conradt and Horvitz, 1999) to an XhoI site; 99 animals from eight independent lines were
scored.
