Fig. S1. Relative phenotypic severity/penetrance in Fgfr1KI embryos. (A) Number of vertebrae caudal to the hindlimb in E14.5-18.5 embryos; each data point represents a single embryo. P values were determined by Student’s t-test comparing the indicated series against *(Fgfr1+/+) or **(Fgfr1wtKI/wtKI, Fgfr1wtKI/Dex4) data. (wtKI homo- and hemizygotes were not significantly different from each other.) (B) Penetrance of defects observed in Fgfr1DFrs/DFrs, Fgfr1wtKI/wtKI and Fgfr1wtKI/Dex4 embryos. n, number embryos phenotyped.
Fig. S2. Responses of immortalized MECs to Fgf. (A, part i) Frs2 IP- 4G10 western blot from immortalized MECs stimulated with 50 ng/ml aFgf. Blot was stripped and re-probed with anti-Frs2 (A, part ii). Arrows indicate the tyrosine phosphorylated Frs2 band detected by both antibodies. Additional bands in A, part ii are probably serine/threonine phosphorylated (upper) and unphosphorylated (lower) forms of Frs2 (Lax et al., 2002). *No antibody IP control. (B) pMAPK and pAkt responses of immortalized MECs stimulated (5¢) with different concentrations of Fgf. RasGAP is a loading control. All stimulations included 5 mg/ml heparin.
