Fig. S1. Schematic representation of a hermaphrodite germline. Shown on top is a cartoon of an adult animal with anterior to the left. The bi-loped gonad occupies much of the central region of the adult. Shown below is a blow-up of the anterior gonad arm. At the most distal end (pink; distal to the uterus) is the distal tip cell (DTC). The DTC induces germ cell proliferation in the adjacent region of the gonad. The central core of each distal arm is anucleate and is occupied by a common cytoplasm known as the rachis. The germ cell nuclei reside on the periphery of the somatic gonad and are not surrounded by a complete plasma membrane. Thus, the germline is a syncytium of nuclei sharing a common cytoplasm. As the germ cells move from the mitotic zone into the transition zone, they enter meiotic prophase. As nuclei approach the bend in the gonad, they cellularize and acquire a complete plasma membrane. Oocyte chromosomes further condense as they progress through diplotene into diakinesis. In diakinesis, the oocytes continue to grow. The oocyte adjacent to the spermatheca (spth) is designated –1 and those farther away are –2, –3, etc. An oocyte from each gonad arm is ovulated into the spth every 20-25 minutes (McCarter et al., 1999) where fertilization occurs. Each gonad arm has a spermatheca containing sperm. Fertilized embryos then exit the spermatheca into a common uterus where they complete their meiotic divisions and first few mitotic divisions before being released into the environment.
Fig. S2. WEE-1.3 depletion causes an increase in the number of oocytes that stain with the pICP-1 antibody. (A-D) Wild-type (WT; A,B) and WEE-1.3-depleted (C,D) animals stained with the pICP-1 antibody (A,C) and TOTO-3 (B,D). Scale bar: 20 mm. (E) Quantitation of pICP-1-positive nuclei by oocyte position in the proximal gonad. Below the graph is a table showing the number of oocytes analyzed.
