Fig. S1. Analysis of
neural crest and sclerotomal contributions to the dorsal aorta. (A)
Co-immunohistochemistry on sections of an E10.5 embryo at the interlimb level
from a ROSA26R mouse line that has been
crossed with a transgenic line expressing a neural crest-specific Cre
recombinase, Ht-PA-Cre (see Materials
and methods). Neural crest derivatives, dorsal root ganglia (DRG) and
sympathetic ganglia (SG) are stained with a b-gal antibody (green);
the smooth muscle cells of the dorsal aorta (DA), stained with an a-smooth
muscle actin antibody (red), are negative. (B) The same section as in A under higher magnification, as
indicated by the white box and arrow. (C)
Section of a somite from an E9.5 Pax3
mutant embryo that lacks neural crest, hybridized with a Pax1 probe to mark sclerotomal cells. (D) The adjacent section where GFP is revealed by
immunohistochemistry. (E)
Co-localisation of Pax1 transcripts and
GFP. Strong labelling is observed in the dermomyotome (DM) and in Pax1-negative
cells extending more ventrally towards the dorsal aorta (DA). NT, neural tube;
M, myotome. Scale bars: in A, 50 mm; in B, 20 mm; in
C-E, 20 mm.
