Fig. S1. The hindbrain is correctly patterned in β-catenin CKO and cAct embryos. β-catenin CKO (A) and cAct (B) embryos were examined between E8.5 and E8.75 with markers of rhombomeres 3-5. Hoxb1 marks rhombomere 4, Fgf3 marks rhombomeres 5 and 6, while Epha4 and Krox20 mark rhombomeres 3 and 5. No differences can be seen between control and mutant embryos. The Krox20/Dlx5 panel in A is the same as that shown in Fig. 2A.
Fig. S2. Epibranchial placode defects in β-catenin CKO embryos. Ngn2 is expressed in the geniculate, petrosal and nodose placodes. In β-catenin CKO embryos, both the geniculate and petrosal placodes are reduced in size by E9.0, with the petrosal placode disappearing by E9.5.
Fig. S3. Shh expression in β-catenin cAct embryos. Normal and β-catenin cAct E9.5 embryos hybridized with a probe for Shh. No difference can be seen between the two embryos. The broken line indicates the outline of the otocyst in each specimen.
