  Fig. S1. Generation of the En1^flox allele. Gene targeting strategy for generating the En1^flox allele. loxP sites were inserted in the intron at the KpnI site in the 3′-UTR of the second exon. An frt-flanked neo cassette was inserted into the intron. The neo cassette was removed by Flp recombination in mice (Rodriguez et al., 2000). Light blue rectangles represent En1 coding regions; dark blue rectangles represent En1 5′ and 3′ UTRs; red rectangles represent the En homeodomain; pink triangles represent loxP sites; red triangles represent frt sites; thick black lines represent 5′ and 3′ external probes used to identify the mutant allele.
