Fig. S1. Ptc1-expression levels in wild-type and Ftm−/− tissues at stage E11.5. In Ftm−/− neural tube and limb, Ptc1 expression is reduced. The upper panel shows the Hprt normalization. Data have been confirmed in two independent experiments.
Fig. S2. Whole-mount in situ hybridisation at stage E10.5 in wild-type and Ftm−/− embryos. In Ftm−/− embryos (B), Shh expression along the anterioposterior axis is partially interrupted in comparison with wild-type embryos (A). The gaps in Shh expression in Ftm−/− embryos are restricted to the neural tube (dorsal Shh expression; black arrowhead in the enlargement), whereas the expression in the notochord (thin blue line, ventral) is unaffected. The positions of the enlargements are marked by the black boxes.
Fig. S3. Multiple sequence alignment of Ftm proteins. The following sequences were aligned: mouse (Mus musculus), human (Homo sapiens), zebrafish (Danio rerio), lancelet (Branchiostoma floridae), sea urchin (Srongylocentrotus purpuratus), capitella (Capitella sp.), lottia (Lottia gigantea), sea anemone (Nematostella vectensis), hydra (Hydra magnipapillata) and the Ftm-related protein Rpgrip1 (mouse). Sequence identities are marked in grey.
