(A) Essentially identical expression patterns of GFP and Helt in the heterozygous mutant. (B) FACS patterns of GFP in heterozygous and homozygous E12.5 ventral mesencephalons. The GFP+ populations are sorted with about 99% purity. (C) Residual GFP expression in postmitotic neurons. GFP+ cells are detected in the mantle layer and also express the postmitotic neuron marker Lim1 (see Fig. 6).
Fig. S2. Helt does not regulate progenitor domain formation. The expression of Nkx2.2, Nkx6.1 and Hnf3β in the ventricular zone is not altered by exogenously expressed Helt. The postmitotic expression of Nkx2.2 in the m2 domain is dependent on Helt activity (see Fig. 4A), but exogenous Helt cannot ectopically induce Nkx2.2+ neurons.
