Fig. S1. Large numbers of dorsally migrating cells are labelled from CMFDA tracer placements in the ventral telencephalon of stage-14 embryonic turtle forebrain slices. (A) The various sites (MGE, LGE, PSB, CX) of tracer placements are indicated on the GABA-immunostained coronal section of a stage-14 turtle brain. PSB tracer placements were performed at the frontier between the pallium and the subpallium, and cortical tracer placements (CX) into the middle of the dorsal pallium. Since the ventral telencephalon of the E14 turtle consists of a single ridge, presumptive MGE tracer placements were performed in the ventral half of the ridge and LGE placements into the dorsal half. (B-C′) Early (stage 14, B,C) and late (stage 16-18, B′,C′) stage frontal slices were labelled in the PSB and MGE, respectively. After 5 days in culture, the placement of CMFDA-coated bamboo fibers in slices was assessed under phase contrast microscopy (B,B′). Fluorescent microscopy in the same slices (C,C′) reveals the distribution of cells labelled from each site (note the site of the bamboo particle on B and B′ images). From the PSB injection site (C), cells migrated outside the slice onto the surrounding agar. From the MGE injection site (C′), cells migrated dorsally and invaded the V-shaped DVR and the dorsal cortex (DC). Scale bar: 500 μm in B,B′.
