Fig. S1. Time course of dp-ERK, myosin accumulation and trh-lacZ expression in four successive stages in a control tracheal placode prior to apical constriction. dp-ERK (magenta), myosin (green) and trh-lacZ (blue). (A,A′) Roughly corresponding to the onset of dp-ERK expression at the stage shown in Fig. 4A. (B-D′) Corresponding to the stages just before and after the images shown in Fig. 4B, when the myosin-enriched cell boundaries started to appear. Note that outward spread of myosin-enriched cell boundaries (arrows) corresponds to the propagation of dp-ERK. Scale bar: 10 μm.
Fig. S2. Myosin expression in pnt mutants. (A-B′) Although pnt mutants are defective in tracheal invagination, they showed some degree of myosin accumulation in partially formed cellular arcs, suggesting that pnt mutants are hypomorphic in this aspect. Arrows indicate the high myosin-GFP accumulation. Scale bar: 10 μm.
Movie S1. Lateral view of T1-T3 of a sqh-GFP-Moesin embryo at stage 11. Tracheal cells invaginated in the recorded period were colored green in the starting frame (tr 1, tr 2). Four 0.7-μm z-stacks covering the apical side of cells were taken every 2 minutes over a period of 1 hour. Developmental time shown in the lower right corner was estimated based on the assumption that dorsal ectoderm cells re-enter mitosis 60 minutes after the start of invagination.
Movie S2. A myosin-GFP-expressing embryo. Arrows indicate signal concentration in cell rows in three successive waves (blue, green, magenta).
Movie S3. An Egfrf24 embryo labeled with sqh-GFP-moesin. Cells undergoing precocious internalization are indicated in orange and major invagination sites are indicated in pink.
Movie S4. A rhodel1 embryo labeled with sqh-GFP-moesin.
Movie S5. A pntdelta88 embryo labeled with sqh-GFP-moesin.
