Fig. S1. Labelling of the E7.5 mouse node by DiI injection and electroporation of the neural plate with a GFP expression vector. (A) Whole E7.5 embryos shown from the posterior surface a few minutes after injection of DiI into the node. Location of the node is indicated by the black arrow in the embryo on the left side. Notice the similar intensity of DiI labelling in all embryos. Subsequent to culture, the genotype of these embryos was determined, as shown by lettering on the ectoplacental cone. It is clear that embryonic genotype at the Lp locus cannot be discerned from embryonic morphological appearance at this stage. Hence, DiI labelling was performed entirely blind to genotype in this study. White arrow indicates ectopic DiI labelling on the yolk sac of one embryo. White arrowhead points to a silk suture strand, which was inserted through the yolk sac in this experiment to enable individual embryos to be distinguished when cultured together in the same tube. (B) GFP expression (arrow) can be detected in the caudal midline 4 hours after electroporation at E7.5 (right lateral view; hf, head fold). Scale bar: 0.4 mm in A for A and B.
