Control experiment depicting ascending volumes of supernatants (from 0 to 30 µl) of centrifuged conditioned media of SiControl-transfected human brain vascular pericytes (HBVPs) migrating in Boyden Chambers in serum starvation (0.1% FBS) for 6 hours. Volumes of conditioned media supernatants reflect the concentrations of secreted matrix metalloproteinase (MMPs). Note the ascending concentration-dependent effect on proMMP2 gelatinase activity as seen in the gel (above) and quantified by densitometry (beneath). This control experiment showed the sensitivity of gel zymography to detect gelatinase activity in a dose-dependent fashion.
