Each profile includes four cells. Peaks of Fmi/E-cad show positions of apicolateral junctions. x-axis represents position on the proximodistal axis in arbitrary units, proximal to the left and distal to the right. y-axis represents fluorescent intensity in arbitrary units − note that these are not comparable between different genotypes. (A,B) Mwh intensity across wild-type cells, from two different wings, at around the time of prehair initiation. Note that peak Mwh levels are seen towards the proximal edge of the cell, but are not tightly associated with the junctions (as marked by Fmi labelling). Intensity is then graded towards the distal cell edge, with lowest levels at the distal edge where prehair initiation occurs. (C,D) Mwh intensity across fz mutant cells, from two different wings, at around the time of prehair initiation. Cells no longer show peak levels towards the proximal rather than the distal edge; however, levels close to junctions are generally higher than levels towards the centre of the cell, suggesting that Mwh is still preferentially stabilised at cell edges. (E,F) Mwh intensity across stbm mutant cells, from two different wings, at around the time of prehair initiation. Cells no longer show peak levels towards the proximal rather than the distal edge, although as in fz wings, Mwh appears to be preferentially stabilised close to cell edges.
