Fig. S1. RT-PCR analysis of Corin expression in vitro. RNA prepared form freshly isolated versGFP+ cells (F) or after the indicated days in culture were analyzed for Corin mRNA expression using β-actin as a control.
Fig. S2. Hair follicle formation is normal in mice lacking Corin. Immunostaining of P3 frozen skin sections from wild-type and mutant mice with anti-Corin antibody. As initiation of hair follicle morphogenesis occurs in three subsequent waves during embryogenesis, this time difference temporarily leads to the presence of three distinct stages of hair follicles that can be easily distinguished in P3 skin and retrospectively referred to the corresponding wave. Note that all three stages of hair follicles can be identified in both wild-type and mutant sections. Also note that a low level of non-specific green staining outside the DP is occasionally observed in both wild-type and mutant sections. Corin, green; Nuclei, red.
Fig. S3. Hair-type distribution in wild-type and mutant Corin mice. No differences in the frequency of the different hair types were observed between wild-type and mutant mice.
Fig. S4. In mice lacking Corin, the majority of zigzag hairs exhibit a yellow tip. (A) Frequency of zigzag hairs with black tip. (B) Frequency of hairs with yellow tip demonstrating that such hairs are unique to mice lacking Corin.
Fig. S5. The extent of pheomelanin deposition in auchene hairs is longer in the absence of Corin. Two representative examples of auchene hairs from wild-type and mutant mice are shown. Higher magnifications of the hair tips are shown in the upper right corner.
Fig. S6. Agouti expression. Agouti transcripts (blue) detected by in situ hybridization to sections of skin harvested at P2 or P3 from FVB (A/A; tyrC/tyrC) mice. At P2 agouti expression is observed in the DP of some awl hair follicles but not the DP of guard hair follicles. Hair bulbs in the boxed regions are shown at higher magnification in the inserts. At P3, agouti expression is also observed in the follicles from the third wave (arrow) and persists in some awl follicles (arrowhead).
