Fig. S1. The Wnt1 promoter and the Pax3 1.6 kb proximal promoter drive Cre expression in neural crest cells with similar kinetics. (A-F) Whole-mount lacZ staining (blue) on E8.5 embryos of the indicated somite (som) number and genotype. The Rosa26R locus has been engineered (R26RlacZ) to express lacZ in cells where Cre is active. Both the proximal Pax3 promoter-driven Cre (Pax3Cre) and Wnt1-driven Cre (Wnt1Cre) are active in rhombomeres R2, R4 and R6 and lineage-mark neural crest cells beginning around the 10-somite stage. Wnt1Cre is active in additional neural crest populations, driving lacZ expression in neural crest cells migrating to the first branchial arch (BA1). C and F are dorsal views of the embryos shown in B and E and demonstrate that both Wnt1Cre and Pax3Cre expression initiates in R6, which provides cardiac neural crest cells, around the 16-somite stage.
