Fig. S1. The strength of the A6 to A5 transformation in F7rF8 depends on the orientation of the Fab-8 fragment. Two male cuticles of F7rF8 homzygotes are shown in dark-field microscopy to visualize the trichome distribution on the cuticles (small hairs). In wild type, the 5th tergite is uniformly covered by trichomes, whereas in the 6th abdominal segment, trichomes are found only on the anterior and ventral sides of the tergite. (A) F7rF8+. The Fab-8 fragment replacing Fab-7 is inserted in the same orientation as it is in its endogenous site, relative to the Abd-B promoter. The trichome distribution on the 6th tergite looks nearly normal, indicating a weaker A6 to A5 transformation of the tergite. (B) F7rF8−. The Fab-8 fragment is inserted in the opposite orientation. In these homozygotes, the trichomes cover most of the 6th tergite, indicating an A6 to A5 transformation.
