  Fig. S1. Hty acts downstream of Ptch1. (A) Ptch1-lacZ is expressed in cells with active Hh signaling in a wild-type E9.5 mouse embryo. (B) An E9.5 Ptch1 mutant embryo exhibits severe neural tube defects and ectopic Ptch1-lacZ expression in all cells. (C) Ptch1-lacZ expression is downregulated in an E9.5 Hty mutant embryo. (D) An E9.5 Ptch1/Hty double-mutant embryo showing partial rescue of the neural tube defect of the Ptch1 mutant and downregulation of Ptch1-lacZ expression. The Ptch1-lacZ expression level is slightly higher in the double mutants than in Hty mutants, which is likely to be due to the hypomorphic nature of the Hty mutation.
