  Fig. S1. Northern blot analysis of PAPSS isoforms in the bm growth plate. Representative northern blot (10 µg of total RNA) of postnatal day 6 cartilage RNA probed for Papss1 and Papss2. Experiments show expression of both isoforms in wild-type growth plates. By contrast, the bm growth plate exhibited normal Papss1 mRNA expression, and marked reduction of Papss2 mRNA expression. Day 6 cartilage was collected and pooled from at least six pups, and a β-actin was used as loading control.

  Fig. S2. Specificity of Ihh interaction with aggrecan. Embryonic day 14 chick cartilage lysates incubated with IHHAP and IHHAPmut in the presence of the aggrecan-specific monoclonal antibody S103L. Significantly more AP activity was immunoprecipitated with IHHAP than with the IHHAPmut protein lacking the Ihh proteoglycan binding domain (P<0.0001, n=3).
