Fig. S1. Characterization of the rabbit anti-L-PGDS antibody. (A) Purified anti-L-PGDS antibody was used (1:500, red) with germ cell marker PECAM (green) in immunofluorescence experiments on XY (left) and XX (right) 12.5 dpc gonad sections. (B) Total extracts from XY and XX 13.5 dpc gonads (dissected without mesonephros) and from HeLa cells transfected with L-Pgds-pcDNA (L-Pgds cDNA) or empty vector (EV) were separated on a 12.5% SDS-PAGE gel; proteins were transferred onto nitrocellulose and submitted to western blots using anti-L-PGDS (1:500, L-Pgds Ab) or anti-SOX9 (1:200, SOX9 Ab) as a control.
Fig. S2. Spatial and temporal expression of L-Pgds in postnatal testis. (A) In situ hybridization for L-Pgds was performed on sections from newborn (top) and adult (bottom) wild-type XY gonads. InT indicates interstitial compartment. (B) Following L-Pgds in situ hybridization, the sections were submitted to immunofluorescence for SOX9 (red) to precisely identify the location of L-Pgds expressing cells (white). Short arrows, Sertoli cells; long arrows, spermatogonia. Testis cords (TC) are highlighted by black and blue dashed lines. Scale bars: 50 or 30 µm in A, 30 µm in B.
