Movie 1. Dynamics of Fascin-GFP in living plasmatocytes. GFP-Fascin was specifically expressed in plasmatocytes (srp-Gal4>UAS-GFP-Fascin) and imaged in live stage 15 embryos. Confocal stills were acquired every 7 seconds and the movie displayed at 5 frames/second for 15 minutes. GFP-Fascin localizes to actin microspikes that polymerize in lamellipodia and display rapid turnover.
Movie 2. Comparison of actin organization in wild-type versus Fascin-deprived living plasmatocytes. Time-lapse confocal analysis of cytoskeletal dynamics in wild-type and snX2 mutant plasmatocytes (srp-Gal4>UAS-GFP-Moe). Images were acquired every 30 seconds and the movie displayed at 5 frames/second for 30 minutes. Lamellipodia of the wild-type plasmatocyte are highly dynamic and polarized towards the direction of migration. By contrast, the lamellipodia of the snX2 plasmatocyte were unpolarized and displayed reduced ruffling.
