Fig. S1. Engrailed represses groove cell behavior cell-autonomously. (A,A′) Misexpression of en with pnr-Gal4 results in loss of the groove marker Odd from the dorsal epidermis despite hh co-expression. (A′) A high-magnification view. Arrows point to cells that have ceased to express Odd owing to the ectopic expression of En. (B,B′) en represses segmental groove formation cell-autonomously (between arrows). (C) Cells that misexpress en and hh (between arrows) do not constrict their apices. Note the flat surface of cells. Stages: 12 (A); 13 (B,C).
Fig. S2. Groove differentiation in slp1, ptc mutants. (A) In slp11 mutant embryos, wg fades, relieving the antagonistic effect of wg on Hh targets. Simultaneous mutation of ptc, which results in maximum strength Hh signaling, leads to the specification of up to 12 rows of groove cells marked by the accumulation of Odd and Ena, as compared with a single row in the wild type. Interestingly, morphologically differentiated grooves form only at the edges of these broad groove cell populations (arrowheads). (B) SEM of an slp11, ptc7 double mutant. Grooves (arrowheads) differentiate only at the interfaces between constricting (arrow) and non-constricting cells. Stages: 13 (A); 12 (B).
Fig. S3. Molecular characterization of odd5. (A,A′) Wild-type Odd protein co-localizes with Sr in the nucleus. (B,B′) In odd5 mutants the truncated Odd protein is distributed diffusely throughout the cell. (C) Schematic representation of wild-type Odd, and the truncated protein in odd5. (D) Distribution of En and Odd in the wild type. (E) In odd5 mutants, segments affected by the pair-rule phenotype have ectopic and partially fused En stripes (asterisks). (F) Distribution of Wg and Odd in the wild type. (G) In odd5 mutants, segments affected by the pair-rule phenotype exhibit an ectopic Wg stripe (asterisks). Stage 12 (A,B,D-F).
Fig. S4. Misexpression of odd-skipped does not induce ectopic groove formation. (A,A′) pnrGal4>UAS-odd embryo. Expression of odd does not cause cell shape changes or Ena accumulation. (B) pnrGal4>UAS-odd embryo. Expression of odd does not cause ectopic groove formation (arrows indicate wild-type grooves). (C) enGal4>UAS-GFP, UAS-hh embryo. Approximately four cell rows posterior to the Hh source are specified as groove cells (arrows). (D,D′) pnrGal4>UAS-odd, UAS-hh embryo. Approximately four cell rows posterior to the Hh source are specified as groove cells (arrows). Stage 13 (A-D).
