Fig. S1. Expression of Ptc1 and Ihh in ShhcreERT2/C embryos suggests redundant Hedgehog signaling in the cloaca. (A) Ptch1 expression in control Shh+/C embryos. (B) Following loss of Shh signaling in ShhcreERT2/C embryos, Ptch1 expression is lost from the genital field (compare red arrowheads) and from mesenchyme surrounding the ventral cloaca, but persists in mesenchyme surrounding the dorsal cloaca (compare black arrows). Loss of Ptch1 from the genital field confirms inactivation of the Hh pathway in ShhcreERT2/C embryos. (C,D) Ihh is expressed in the gut endoderm and the cloaca but not in the genital field in both control (C) and ShhcreERT2/C (D) embryos. Persistence of Ptch1 around the region of Ihh in the cloaca after deletion of Shh suggests that Shh and Ihh are redundant in the cloaca but not in the external genitalia.
Fig. S2. Cloacal and urethral plate epithelial progenitor cells persist in the absence of Shh signaling. Immunohistochemical staining with p63 antibody, which marks epithelial progenitor cells, shows positive staining in the urethral and cloacal epithelium of Shh+/C and ShhcreERT2/C embryos.
