Fig. S1. Distinct spatial and temporal movement behaviors of cells from the AHF/SpM and the differentiated SpM. (A,A′) Injections of DiI and DiO into St. 8 embryos (A). DiI was injected into the AHF/SpM and DiO to the differentiated SpM, as seen in the transverse section (A′). (B-B′′) After 8 hours of incubation at St. 10 (B), the DiI is seen in the splanchnic mesoderm dorsal to the outflow tract (B′), whereas the DiO is seen in the myocardium of the linear heart tube (B′′). (C-C′′) After 16 hours of incubation at St. 12 (C) the DiI is detected in the outflow tract myocardium (C′), whereas the DiO is seen in the future ventricle (C′′) (n=10). AHF/SpM, anterior heart field/splanchnic mesoderm; ph, pharynx; hrt, heart; nt, neural tube; oft, outflow tract.
Fig. S2. The molecular division within the myogenic core of the first branchial arch. (A-G) The molecular signature of the mesoderm core of BA1 (schematically depicted in G) was assessed by immunofluorescence for Isl1, Myf5 and Nkx2.5 (A-F). At St. 18, a transverse section through BA1 was stained with DAPI (A,D), for Nkx2.5 (B, blue), Isl1 (C, red) or Myf5 (E, green). The merged image (F) demonstrates that while Isl1 and Nkx2.5 are expressed in the distal part of BA1 mesodermal core, Myf5 is localized more proximally. Cells within the mesodermal core (enclosed by a dashed ellipse) are seen to be enveloped by neural crest cells. nc, neural crest; nt, neural tube; BA1, branchial arch 1; mes, mesoderm; hrt, heart; ph, pharynx; ecto, ectoderm.
