Movie 1. Cell rearrangements leading to the presence of a ptc-expressing cell within the en domain. A single ptc-expressing cell (green) becomes isolated from the rest of the A compartment by the dorsalward migration of en cells (red) to the leading edge during DC. This cell rearrangement reproducibly occurs in segments A1-A5.
Movie 2. Specific binding of filopodia to matching cells during DC. Movie shows zippering of part of the DC leading edge. During zippering, filopodial interactions occur between cells of the same colour but not between cells of different colours, resulting in perfect alignment of the epithelial sheets following fusion.
Movie 3. Realignment of misaligned epithelial sheets by filopodial searching and pulling. Movie showing DC occurring in an embryo in which the two epithelial sheets are initially out of alignment. However, long-range interactions occur between filopodia of matching cells: initially between en cells (red), then later between ptc cells (green), and these gradually pull the sheets back into correct alignment.
Movie 4. Matching in an embryo with asymmetries between the opposing epithelial sheets. Movie showing DC occurring in an embryo in which the two epithelial sheets are patterned slightly differently, resulting in a fusion between ptc cells (green) that are not in equivalent positions. This blocks matching of the adjacent en cells (red) and these blocked cells subsequently fuse with opposing en cells from the neighbouring segment, demonstrating that matching is not segment-specific.
Movie 5. Filopodial matching during wound healing. A laser wound was made in the ventral epithelium of an embryo across a stripe of en cells (red). During healing of this wound, filopodial interactions are observed only between matching cells, initially between ptc cells (green), then later between en cells. Finally, fusion of en cells results in the regeneration of the en stripe.
