Movie 1. Neural crest cells in the presence of ephrinB1-Fc fusion protein do not sort into discrete sympathetic ganglia. HH St.10 chick embryos injected with the GFP-encoding plasmid pMES label pre-migratory neural crest cells. At E3, when neural crest cells are dispersed adjacent to the dorsal aorta in the anterior and posterior directions, ephrinB1-Fc (400 ng/ml) was injected lateral (bolus measuring one somite in length) to the GFP-expressing neural crest cells. At E3.5, after normal sorting of sympathetic ganglia into discrete ganglia should have occurred, a trunk sagittal explant was cultured and fluorescently-labeled neural crest cell movements were imaged with time-lapse confocal microscopy. Images were collected every 2 minutes using a 20× objective, zoom=3.
