The transgene contains ∼ 5.4 kb upstream of the initiation codon, exons 1-4 and adjacent introns 1-4, and part of the 5′ region of exon 5 of rat nestin. Whole mount section (B) or cross section (C) through the neural tube of an X-gal stained Nestin-CreERT2;R26R-lacZ embryo treated with tamoxifen at E10.5 and harvested 24 hours later at E11.5.

(A,A′) X-gal staining in whole-mount and vibratome sections of a second line of transgenic mice (B2C-8) carrying the Ascl1-CreERTM transgene show expression of the lacZ reporter in a similar pattern but at different levels than that shown in Fig. 3C,C′ when induced with tamoxifen at E11.5 and harvested at E12.5. Treatment with tamoxifen at E15.5 results in labeling of cells in the gray and white matter by P30 similar to that shown in Fig. 4C.
