      Fig. S1. At E9.5, the Otx2/Gbx2 expression boundary is shifted posteriorly. In situ hybridization showing the expression of Otx2 and Gbx2 on consecutive sagittal sections of E9.5 control and mutant embryos, and using digoxigenin-labeled probes, as indicated. The isthmic constriction is indicated in the mutant by an arrow (G). The anterior expression border of Gbx2 in the control is indicated by an arrow and is clearly on the anterior side of the isthmic constriction (C). In the mutant (E,G), this border is posterior to the isthmic constriction. In the mutant (F,H), the posterior expression boundary of Otx2 is also shifted posteriorly (arrowhead in H) when compared with the control (arrowhead in D).

      Fig. S2. In situ hybridization showing the expression of Fgf8 and Otx2 on consecutive sagittal sections of E9.7 control and mutant embryos, and using digoxigenin labeled probes, as indicated. At this stage of development, the posterior border of Otx2 and the anterior border of Fgf8 retain a normal boundary relationship being not overlapping. However, relative to the isthmic constriction, which is indicated by an arrow in the control (C) and in the mutant (E), it appears that the area including the posterior border of Otx2 and the anterior border of Fgf8 is posteriorly shifted in the mutant. In addition, the patchy expression of Fgf8 and Otx2 is evident as this stage of development.
