Fig. S1. 35S::CLE41 and 35S::CLE42 phenotypes are pxy dependent. (A-J) Diagrammatic representation of pxy (A) and 35S::CLE42 (B) vascular tissue. Toluidine Blue sections of inflorescence stem vascular tissue from pxy (C), 35S::CLE42 (D), pxy 35S::CLE41 (E) and pxy 35S::CLE42 (F) demonstrate that CLE42 overexpression phenotypes are similar to those of 35S::CLE41 (see Fig. 4B) and that in both cases these phenotypes are dependent on a functional copy of PXY. This is also the case in hypocotyls (G-J), where CLE41/42 overexpression phenotypes (H) are suppressed to the level of pxy mutants (I,J). Intercalation of vascular tissue is common to pxy (K,M) and 35S::CLE42 (L,N). White arrowheads show phloem surrounded by xylem in Aniline Blue-stained sections. Ph, phloem; X, xylem. Scale bars: 50 µm in C-F,M,N; 100 µm in G-L.

Fig. S2. Comparison of 35S::CLE41, IRX3::CLE41 and SUC2::CLE41 lines with equivalent levels of CLE41 expression in stems. (A) qPCR normalised to ACT2 showing relative CLE41 expression levels in wild-type, 35S::CLE41, IRX3::CLE41 and SUC2::CLE41 lines shown in the figure. (B-E) Toluidine Blue-stained hand sections of vascular bundles in inflorescence stems of wild-type (B), 35S::CLE41 (C), IRX3::CLE41 (D) and SUC2::CLE41 (E) lines. Arrowheads point to mis-oriented cell divisions. (F-I) Analine Blue-stained hypocotyl hand sections show that vascular tissue is disrupted in 35S::CLE41 (G) and IRX3::CLE41 (H) lines compared with wild-type (F) and SUC2::CLE41 (I) lines. Scale bars: 30 µm in B-E; 50 µm in F-I.
